Portland has a small airport, from which you can reach Downtown via rental car, taxi, or bus. Cab fare is usually $20 and is probably the way to go as you are unlikely to need a rental car after arriving downtown.

Usually a cheaper option and because Boston can be reached on a direct flight from most locations, is not much more time consuming. From Boston, you can get to Portland via rental car, coach, or train. Trip will take 1.5 to 2 hours in a rental car. The coach is operated by Concord Coach Lines (2-2.5hrs, $47 roundtrip). Train is operated by Amtrak (2.5hrs, $40-70 roundtrip).

Also an option, but not a good one.
